Abstract
Type IV collagenase (including MMP-2 and MMP-9) is closely related to tumor invasion, metastasis and malignant progression. By constructing a eukaryotic expression vector that can express the anti-type IV collagenase scFvM97 antibody gene in the endoplasmic reticulum of eukaryotic cells, Gene therapy was performed on human highly metastatic giant cell carcinoma PG cell line using lipofectamine method. The results showed that intracellular expression of scFvM97 had a significant inhibitory effect on the growth and in vitro invasion ability of PG cells secreting type IV collagenase. , and can significantly reduce the tumorigenicity of PG cells in nude mice and prolong the survival time of animals. The above results suggest that intracellular antibody technology can fundamentally inhibit the activity of type IV collagenase in the key step of protein processing and secretion, and can be used in tumor gene therapy. has important application prospects.
