Application Of Human Amniotic Membrane Stromal Cells As Seed Cells For Bone Tissue Engineering

Abstract
? Purpose: To initially explore the potential of human amniotic membrane stromal cells (hamc) as seed cells for bone tissue engineering. Methods: hamc was isolated and cultured, and the third generation hamc was induced into osteogenesis in DMEM culture medium containing dexamethasone (0.1 ¦Ìmol/l), vitamin C (50 mg/l) and ¦Â-glycerol sodium phosphate (10 mmol/l). After culturing for 1 week, alizarin red staining was performed to count the number of calcified nodules, and immunofluorescence cytochemical staining was performed to detect the expression of type I collagen and alkaline phosphatase. Immunohistochemical staining was used to detect the expression of fasl in amniotic membrane and hamc. Results: After osteogenic induction of hamc, obvious calcified nodules were formed, with an average of 18 per well. The cells in the calcified nodules expressed positive expression of type I collagen and alkaline phosphatase. The positive expression of fasl can be detected in cells in human amniotic membrane tissue and in hamc cultured in vitro. Conclusion: hamc has the potential to become an ideal seed cell for bone tissue engineering.
