Abstract
The full-length cDNA sequence of the ¦Á1 gene (col1a1) encoding grass carp muscle type I collagen was cloned for the first time using PCR and race methods. It is 5772 bp, and its open reading frame is 4347 bp, encoding 1448 amino acids. The results of blast homology analysis showed that the amino acid sequence of grass carp col1a1 gene has high homology with zebrafish and goldfish, 93.90% and 93.60% respectively, showing high conservation. Phylogenetic tree analysis shows that this gene is in the same branch as zebrafish and goldfish, and has the closest relationship. Bioinformatics analysis shows that the relative molecular mass of grass carp col1a1 protein is 137.2ku, the theoretical isoelectric point is 5.44, and it is an ¦Á-helix ¦Â-sheet triple helix structure protein. There are 18 triple helical repeat domains, 22 low-complexity domains, and 17 functional domains. The col1a1 protein has two calcium-binding regions 33~69 and 1249~1355 and one zinc-binding region 62~104. The tissue expression results detected by the semi-quantitative RT-PCR method showed that the col1a1 gene was expressed in eight tissues of grass carp muscle, intestine, hepatopancreas, gills, skin, fin rays, kidneys and spleen, among which the col1a1 gene was expressed in the skin, gills The mRNA expression levels in , kidney, and fin ray tissues were higher than those in the other four tissues (p<0.05).
