Abstract
Objective To establish a method for determining the relative molecular weight distribution of small molecule oligopeptides in fish skin collagen hydrolyzate. Method: High-performance gel exclusion chromatography was used with three TSK-GEL G2000SWXL columns (300 mm¡Á7.8 mm, 5 ¦Ìm) connected in series at room temperature; the mobile phase was acetonitrile-0.1% trifluoroacetic acid solution (15:85), flow rate 0.6 mL?min-1, detection wavelength 220 nm. Results: Within the relative molecular weight range of 149 to 2 490 Da, the logarithm of the relative molecular weight of the oligopeptide had a good linear relationship with its retention time (r=0.995 6); both precision and repeatability RSD were <0.1%. Conclusion This method is simple, accurate and reproducible, and can be used as a method to determine the molecular weight distribution of small oligopeptides.
