Abstract
Objective: To explore the effect of Kangganxian Chongji on the expression of type ¢ñ collagen (Col¢ñ) protein in liver tissue of mice with liver fibrosis caused by immune damage caused by pig serum, and to clarify the therapeutic effect of Kangganxian Chongqing on mice with liver fibrosis. Methods: 75 female Balb/c mice were divided into 5 groups, with 15 mice in each group: normal control group, liver fibrosis model group, colchicine group, low-dose Kangganxian Chong group, and high-dose Kangganxian Chong group. Group. Except for the normal control group, the other groups used intraperitoneal injection of pig serum to prepare liver fibrosis mouse models. While modeling, mice were treated with drugs for 7 weeks. At the end of the 7th week, the mice were sacrificed, and the liver tissues were collected, fixed, sectioned, HE stained, and observed under a light microscope. Other liver tissues were used for real-time quantitative PCR and Western blotting detection. Results: Liver histopathology results showed that compared with the model group, the inflammation in the liver tissue was reduced in the Kanggan Chongqing treatment group; compared with the liver fibrosis model group in the low-dose Kangganxian Chongqing group, the mRNA expression level of Col¢ña2 in the liver tissue was significantly higher. Reduced (P<0.01); Western blotting test results showed that the expression of ColIa1 in the liver tissue of the high-dose Kangganxian Chongji group was also significantly reduced. Conclusion: Kangganxian Chongqing can significantly reduce the expression of Col¢ñ in the liver tissue of mice with immune-damaged liver fibrosis, suggesting that Kangganxian Chongqing has a significant therapeutic effect on mice with liver fibrosis.
