Abstract
In order to reduce the mass transfer limitations during the in vitro construction of engineered tissues, a self-developed perfusion bioreactor system was used to construct engineered dermis using human skin fibroblasts as model cells and PET (Poly(ethyleneterephthalate)) as a three-dimensional scaffold. tissue to examine the effect of perfusion rate on fibroblast growth and extracellular matrix synthesis. Human skin fibroblasts were perfused and seeded on PET scaffolds, and cultured at perfusion rates of 0.02, 0.2, and 0.8 cm/min for 18 days. The results show that perfusion culture can promote cell proliferation and improve the uniform distribution of cells in the 3D scaffold; compared with static culture and a perfusion rate of 0.02cm/min, culture at a perfusion rate of 0.2cm/min and 0.8cm/min, It increases the glucose specific consumption rate of cells and stimulates cells to synthesize and secrete more collagen matrix, but increases the proportion of collagen lost to the culture medium. Perfusion culture is an effective method for constructing engineered dermal tissue in vitro.
