Abstract
Fed-batch fermentation was used to study the effects of four different intermittent cycles of nitrogen supplementation on cell growth and expression of human-like collagen, as well as changes in cell plasmid stability, cell colony formation ability and acetic acid production during the fermentation process. . The results show that under the cyclic operation of continuously adding 36 ml of nitrogen solution at 10-min intervals (completed in 22 s), the stability of cell plasmids was significantly improved, the ability of cell colony formation decreased slightly, and the amount of acetic acid generated was low, only 0.7 g/l, the final cell density is 84g/l, and the expression of human-like collagen can reach 14g/l, which is the highest among the 4 operating methods.
