Effects Of Estrogen On Expression Of Type I Collagen And Matrix Metalloproteinase Activity In Bone Of Ovariectomized Rats

Abstract
In order to explore the molecular mechanism of abnormal collagen metabolism in postmenopausal osteoporosis, RT-PCR and GEL-SDS-polyacrylamide gel electrophoresis (PAGE) were used to detect the mRNA level of type I collagen and matrix metalloproteinase (mRNA) in the bone tissue of ovariectomized rats, respectively. matrix metalloproteinase (mmps) activity, and the amount of type I collagen in bone tissue was observed by immunohistochemistry. The mRNA expression of type I collagen in bone tissue of ovariectomized rats decreased by about 26.3%, and the activity of pro-mmp-9 was significantly increased (p<0.05). Application After estrogen treatment, the mRNA expression of type I collagen increased by 34.1% compared with the ovariectomy group, and the increase in pro-mmp-9 activity was significantly reduced. The activity of mmp-9 gelatinase in bone tissue was significantly increased after ovariectomy, and the mmp-9 gelatinase activity in bone tissue was significantly increased after estrogen treatment. The significant decrease in enzyme activity (p<0.05) indicates that the decrease in bone type I collagen mRNA expression and the increase in pro-mmp-9 activity are one of the mechanisms of postmenopausal osteoporosis.
