Effects Of Integrin ¦¡2¦¢1 On Adhesion And Chemotactic Behavior Of Liver Cancer Cells

Abstract

Micropipette technology was used to study the effect of integrin ¦Á2¦Â1 on the adhesion characteristics of hepatocellular carcinoma (HCC) cells SMMC-7721 and type IV collagen lining surfaces and the chemotactic behavior of type IV collagen of the cells. Observe the effects of Anti-¦Á2 and Anti-¦Â1 on the adhesion of SMMC-7721 cells to type IV collagen-coated surfaces; use the double micropipette experiment method to observe the pseudopod formation process of SMMC-7721 cells and the effects of Anti-¦Á2, Anti- Effect of ¦Â1 on pseudopod formation in SMMC-7721 cells. Flow cytometry was used to analyze the expression of integrin ¦Á2 and ¦Â1 subunits on the surface of SMMC-7721 cells. The results showed that the adhesion force between SMMC-7721 cells and the surface lined with 5 ¦Ìg/mL type IV collagen was 952¡À134 (¡Á10^-10N, n=60). The adhesion force decreased to 605¡À128 (¡Á10^-10N, n=60); when adding 10¦Ìg/mL Anti-¦Á2, the adhesion force decreased to 579¡À57 (¡Á10^-10N, n=50). The adhesion force decreased to 449¡À119 (¡Á10^-10N, n=60) when 5 ¦Ìg/mL Anti-¦Â1 was added; the adhesion force decreased to 220¡À78 (¡Á10^-10N, n=60) when 10 ¦Ìg/mL Anti-¦Â1 was added. n=55). The double micropipette chemotaxis experiment showed that when the same concentration of type IV collagen was added to both sides of the micropipette, pseudopodia were formed in the cells on both sides of the micropipette. On this basis, Anti-¦Á2 and Anti-¦Â1 were added to one side of the micropipette respectively. , the pseudopod growth curve of SMMC-7721 cells showed obvious inhibition, and compared with the opposite side where the antibody was not added to the side of the micropipette, the pseudopodia of the cells on this side increased significantly. Flow cytometry analysis showed that the expression rates of integrin ¦Á2 and ¦Â1 subunits in the cells studied were 23.17% and 95.78% respectively. The above results indicate that integrin ¦Á2¦Â1 is an important receptor basis for the adhesion of SMMC-7721 liver cancer cells to type IV collagen-coated surfaces and the formation of chemotactic pseudopodia of SMMC-7721 cells for type IV collagen.
