Effects Of Mutant Human Hgf (Tvnk1) On Ccl4-Induced Liver Fibrosis In Rats

Abstract
Objective: To explore the effects of mutant human hepatocyte growth factors (HGFK132E, R134E, tvNK1) on carbon tetrachloride (CCl4)-induced liver fibrosis in SD rats. Methods: tvNK1 was prepared in large quantities through biological fermentation and injected intraperitoneally into SD rats with CCl4-induced fibrosis. Liver tissue was collected 6 weeks later, and the effect of tvNK1 on type I fibrosis in the liver of SD rats was detected by real-time PCR and Western blot. Effects on the expression of collagen (Collagen type I, Col1A1), type IV collagen (Collagen type IV, Col4A1) and ¦Á-smooth muscle actin (¦Á-SMA) at the mRNA and protein levels, and further The effects of tvNK1 on liver morphology and liver tissue collagen fibers of fibrotic SD rats were observed by HE and Masson staining. Results: SDS-PAGE test results showed that tvNK1 with a purity of ¡Ý95% was obtained. Real-time PCR and Western blot results showed that tvNK1 reduced the expression of Col1A1, Col4A1 and ¦Á-SMA at the mRNA and protein levels in the liver of fibrotic rats. HE and Masson staining results showed that tvNK1 slowed down the pathological changes in liver structure of fibrotic rats and reduced the content of collagen fibers in the liver. Conclusion: tvNK1 can inhibit CCl4-induced liver fibrosis in rats and provide experimental basis for preventing liver fibrosis and other organ fibrosis diseases.
