Abstract
Objective: To observe the effect of sodium tanshinone II Asulfonate (STS) on angiotensin II (angiotensin II, Ang II)-induced collagen synthesis and TGF-¦Â1 activation in atrial fibroblasts. Methods: Neonatal rat atrial fibroblasts were cultured, collagen content was detected, and collagen synthesis rate was measured by [3H]-proline incorporation method as an indicator of myocardial fibrosis. ELISA method was used to detect the content of active TGF-¦Â1 and total TGF-¦Â1 in the cultured cell supernatant. Westernblot determined the expression of thrombospondin-1 (TSP-1). Results: AngII could significantly increase the collagen content and collagen synthesis rate of atrial fibroblasts; TSP-1 expression and active TGF-¦Â1 secretion were increased, while total TGF-¦Â1 expression had no significant change. After STS treatment, except for total TGF-¦Â1, other indicators were significantly down-regulated. Conclusion: STS can reduce Ang II-induced collagen secretion and synthesis rate in atrial fibroblasts, and the mechanism is related to inhibiting the TSP-1/TGF-¦Â1 pathway.
