Effects Of Ultra-Early Hyperbaric Oxygen Therapy On Femoral Bone Calcium, Bone Biomechanics And Collagen In Rats With Total Spinal Cord Transection Injury¢Ù

Abstract

Abstract: Objective To explore the effects of ultra-early hyperbaric oxygen therapy on femoral bone calcium, bone biomechanics and collagen in rats with total spinal cord transection injury. Methods 75 Sprague-Dawley rats were randomly divided into sham operation group (n=15), model group (n=20) and hyperbaric oxygen group (n=40). The hyperbaric oxygen group was further divided into 3-hour group (hyperbaric oxygen group). Group 1, n=20) and 12-hour group (Hyperbaric Oxygen Group 2, n=20). The sham operation group only underwent laminectomy, while the other three groups all underwent complete transverse transection of the spinal cord at the T10 level. Hyperbaric oxygen group 1 and hyperbaric oxygen group 2 started hyperbaric oxygen therapy at 3 and 12 hours after surgery respectively, with 10 days as one course of treatment, totaling 3 courses. After treatment, the right femurs of each group were taken to measure femoral biomechanical performance parameters, bone calcium and bone hydroxyproline (Hyp) content; HE staining and Masson staining were used to observe the morphological changes of bone trabeculae and collagen. Results After treatment, compared with the sham operation group, the femoral structural mechanics and material mechanics parameters of the model group decreased (P<0.05), and the bone dry weight, bone calcium and Hyp content decreased (P<0.05); the number of trabeculae and collagen decreased, Sparse, broken, and poorly arranged. Compared with the model group and the hyperbaric oxygen group 2, the structural mechanics and material mechanics parameters of the femur in the hyperbaric oxygen group 1 increased, the bone dry weight, bone calcium and Hyp content increased (P<0.05), the number of bone trabeculae and collagen increased, and the continuity Better and neatly arranged. Conclusion Ultra-early hyperbaric oxygen therapy can significantly improve the femoral bone calcium content, collagen content and quality, and biomechanical properties of rats with total spinal cord transection injury.
