Experimental Study On Long-Term Outcomes Of Porcine Collagen Membrane Xenograft Subcutaneous Transplantation

Abstract

Objective To explore the long-term outcomes of porcine collagen membrane (trade name: sterile biological wound protection membrane) after implantation in rats. Methods Twenty SD rats were randomly divided into two groups, with 10 rats in each group. The rats in the experimental group were subcutaneously implanted with porcine collagen membranes on their backs, while those in the control group were not implanted with porcine collagen membranes. In the experimental group, samples were taken continuously at 3, 7, 15 days and 1, 3, 6, and 12 months after surgery, and gross and histological observation of inflammatory reactions, CD31 immunohistochemical staining to observe neovascularization, and transmission electron microscopy to observe collagen arrangement. ; In the control group, the skin tissue from the same part of the rat’s back was taken for observation and comparison 12 months after surgery. Results The postoperative incision of the rats in the experimental group healed well without obvious redness, swelling and inflammatory reaction. Gross observation showed that as time went by, the porcine collagen membrane was in close contact with the surrounding tissue, without obvious rejection, and the porcine collagen membrane was still clearly identifiable at 12 months. Histological observation showed that infiltration of fibroblasts and inflammatory cells could be seen in the experimental group 7 days after surgery; obvious proliferation of microvessels and fibroblasts could be seen at the connection with the host tissue on 15 days and 1 month, and the inflammatory reaction gradually subsided; 3 months At 6 and 12 months, there are abundant blood vessels and cells around the porcine collagen membrane, and blood vessels and cells can be seen inside; at 6 and 12 months, the proliferation of blood vessels and fibroblasts in the porcine collagen membrane gradually decrease, new collagen increases, and the edges are fused. The number of inflammatory cells in the experimental group at 15 days and 1 month after surgery was significantly higher than that in the control group at 12 months after surgery (P<0.05); compared with the control group at 12 months, the difference was not statistically significant (P>0.05). CD31 immunohistochemistry staining showed that intramembranous angiogenesis was visible in the experimental group 15 days after surgery, intramembranous and extramembranous angiogenesis was obvious at 1 month, and angiogenesis was significantly reduced at 12 months. Compared with the control group, the positive expression of CD31 in the experimental group was significantly reduced 15 days and 12 months after surgery, and the positive expression of CD31 was significantly increased 1 month after surgery, and the differences were statistically significant (P<0.05). Transmission electron microscopy showed that the collagen arrangement of the grafts in the experimental group did not change significantly 12 months after surgery compared with before implantation, and pericytes and fibroblasts were visible inside. Conclusion: The long-term structure of porcine collagen membrane after implantation in rats is stable and has good immune tolerance properties.
