Abstract
In this study, type ¢ò collagen (C¢ò) from Agkistrodon japonicus was extracted by restriction pepsin enzymatic hydrolysis method, and purified using ion exchange chromatography. The obtained protein was analyzed by SDS-PAGE gel electrophoresis, ultraviolet spectrophotometry, and infrared spectroscopy. Absorption spectroscopy and mass spectrometry were used for characterization and identification. The results showed that the relative molecular mass of Akiformis C¢ò is about 130kDa, and its ultraviolet absorption peak is about 223nm. The infrared spectrum has characteristic absorption at 1200~1450cm-1, indicating that Akiformis C¢ò has The characteristic sequence Gly-XY peptide segment has a triple helix structure. By analyzing and comparing the protein peptide mass spectra of Aki snake C¢ò and bovine C¢ò, it was found that they have similar peptide segments and C¢ò structural characteristics. The above identification and characterization results prove that the protein extracted by enzymatic hydrolysis is Akidonia C¢ò, which can provide an experimental basis for further pharmacological research on Akidonia medicinal material C¢ò.
