Abstract
In recent years, collagen has received more and more attention in biomaterial applications due to its good biological properties. In order to establish a fast and efficient purification method of turtle-derived collagen and explore its application value in biomaterials, first Van Gieson staining and picric acid-Sirius red staining were used to observe the structure of collagen fibers in the skirt. It was found that the content of collagen fibers in the skirt was very high and the type was mainly type I. Dialysis bags with different molecular weight cutoffs were used to directly dialyze and purify the skirt collagen crude extract. It was found that the dialysis bag with a molecular weight cutoff of 100 kDa had the best purification effect on skirt collagen after 48 hours of dialysis. SDS-PAGE test showed that Almost no stray tape. The biological properties of skirt collagen, including water absorption and in vitro degradability, were investigated. Its water absorption and water holding capacity were as high as 12.06 g/g and 98.21% respectively, and it was completely degraded after 72 hours; on the skirt collagen sponge Hemolysis, skin sensitization, liver wound hemostasis, and wound skin healing were studied, and compared with cross-linked collagen. It was found that neither skirt collagen nor cross-linked collagen would cause hemolysis and skin damage in SD rats. Allergy, both have good hemostatic effect, and skirt collagen can significantly shorten the healing time of wound skin and has good ability to promote wound skin healing, while cross-linked collagen has poor effect. This study shows that skirt collagen exhibits excellent biological properties and has great application value in the field of biomaterials.
