Abstract
In order to clarify the relationship between the nematode cuticle collagen gene (Col) and root-knot nematode diseases, specific primers were designed using the predicted Col from the nematode genome, and the Meloidogyne incognitacol gene MiCol of Meloidogyne incognita was cloned. The complete coding region of this gene is 903bp long and encodes 300 amino acids. The sequence identity of the cloned MiCol and the predicted MiCol gene is as high as 99.67%, and the identity of the amino acid sequence is as high as 100%. Using virus-mediated gene silencing technology, it was introduced into tomato plants and inoculated with Meloidogyne incognita. After 60 days, the number of root knots of tomato plants treated with the silencing vector pTV-MiColi was reduced by 49.4% and 49.2% respectively compared with the empty vector control and clear water control. , indicating that MiCol gene silencing significantly reduced the infection number of Meloidogyne incognita.
