Optimization Of Cod Skin Collagen Peptide Chelated Iron Process Using Response Surface Methodology

Abstract
Response surface methodology was used to optimize the conditions for the chelation reaction between cod skin collagen peptide and ferrous chloride to prepare small molecule peptide chelated iron products. A quadratic orthogonal rotation combination test was conducted at 5 levels using three factors: pH value, mass ratio of small molecule peptide to FeCl2, and mass fraction of small molecule peptide liquid, and a quadratic regression equation for chelate yield was established. The results show that the optimal chelation process conditions are the mass ratio of collagen peptides to ferrous chloride 4:1, the mass fraction of small molecule peptide liquid 3.5%, and pH 7.0. Under these conditions, the chelate yield was 37.31%, which was close to the model¡¯s predicted value of 37.46%. Infrared spectrum detection results show that ferrous ions are chelated with NH2+ and COO- in small molecule peptides, which is a new type of chelate.
