Optimization Of Extraction Conditions Of East Zhejiang White Goose Skin Collagen Using Response Surface Method

Abstract
Taking East Zhejiang white goose skin as the object, the impact of acid type, pH value, solid-liquid ratio and extraction time on collagen extraction rate was explored. The type of acid was first determined, and then based on single factor experimental analysis, response surface optimization was used. Extraction process. The results show that the optimal extraction process is acid type lactic acid, pH 1.9, solid-liquid ratio 1:50 (g/mL), and extraction time 60 h. Under these conditions, the extraction rate of goose skin collagen reached 30.12%, and the relative deviation compared with the theoretical prediction value was 0.13%. It is deduced from UV spectrum that it is a typical type ¢ñ collagen, and infrared spectrum analysis can show that it has a complete triple helix structure.
