Preliminary Study On The Use Of Dynamic Culture Of Thermosensitive Collagen To Construct Tissue Engineering Complexes

Abstract

Objective To study the cell-scaffold complex constructed in vitro using BMSCs as seed cells, thermosensitive collagen hydrogel (TCH) as cell adhesion matrix and poly-L-lactic acid (PLLA) as scaffold material under dynamic culture conditions. Morphological and functional characteristics. Methods BMSCs derived from the long bones of Fischer344 rats were isolated and expanded to the third generation in vitro as seed cells. PLLA fiber woven materials were used as scaffolds and TCH was used as the cell adhesion matrix to construct cell-scaffold complexes. The cell-scaffold complexes in the experimental group and the control group were cultured dynamically and statically respectively. After 7 days of in vitro culture, gross observation and scanning electron microscopy of the two groups of complexes were performed, and the total DNA content in the complexes after 0, 1, 3, and 7 days of culture was detected. Results After culture in both ways, collagen surrounded the PLLA scaffold material and formed a dense membrane structure. Scanning electron microscopy observation showed that the BMSCs on the surface of the complex in the experimental group were spindle-shaped and neatly arranged along the long axis, with good cell activity. Villus bulges were visible on the surface. In the longitudinal section, the PLLA fibers were filled with collagen fibers and cells. The BMSCs on the surface of the complex in the control group showed In the extended state, there are relatively few cells between PLLA. The total DNA content in the cell-scaffold complex in the experimental group was significantly higher than that in the control group at 1, 3, and 7 days of in vitro culture (P < 0.05). Compared with the 0th day of culture, the total DNA content of the two groups on the 1st, 3rd, and 7th day of culture gradually increased, but only the total DNA content on the 7th day was statistically different from that on the 0th day of culture (P < 0.05). Conclusion The composite extracellular matrix constructed by combining TCH and PLLA fibers has good biocompatibility. Dynamic culture can promote the distribution of BMSCs on the surface and inside the material, promote cell proliferation, and can be used for the in vitro construction of tissue engineering complexes.
