Prokaryotic Expression And Functional Study Of A New Type Of Recombinant Human Collagen That Promotes Cell Adhesion

Abstract
Select a gene sequence that is homologous to the corresponding sequence of the COL1A1 gene, is suitable for expression in E. coli, and contains codons encoding the GER tripeptide that promotes cell adhesion. The sequence was amplified from human tissue using RT-PCR technology, and the pET28a(+)-COL recombinant expression plasmid was constructed and transformed into E. coli BL21(DE3)pLysS. IPTG induced expression to obtain recombinant human collagen RHDC, and the expression amount reached about 40% of the total bacterial protein. SDS-PAGE and Western Blot results suggested that the collagen has a triple helix structure. Cell adhesion tests prove that RHDC has the ability to promote cell adhesion and has the potential to be used in the field of biomaterials
