Protective Effect Of Rapamycin On Diabetic Nephropathy Rats

Abstract
Purpose: This experiment mainly studies the protective effect and possible mechanism of rapamycin (rapa) on the kidneys of diabetic rats. Methods: 36 rats were randomly divided into 3 groups: normal control group: 12 rats; diabetic group: 12 rats; diabetic rapamycin treatment group: 12 rats. Diabetic rat model was established by intraperitoneal injection of streptozotocin. Detect changes in biochemical indicators of rats, perform histological examination, observe pathological changes in renal tissue, and use westhernblot method to detect the levels of proliferating cell nuclear antigen (pcna) and type ¢¤ collagen (col-¢¤) protein in renal tissue. Results: Compared with the control group, glu, scr, bun, and upro were significantly increased in the rats in the diabetic group. The rat kidneys were significantly enlarged, the glomeruli were hypertrophied, the thickness of the capillary basement membrane was significantly increased, and the renal cortex pcna and col-¢¤ were significantly increased. protein expression was significantly increased. After rapamycin treatment, all the above indicators except blood sugar were partially suppressed. Conclusion: Rapamycin can reduce renal hypertrophy in diabetic rats, inhibit mesangial extramatrix aggregation, downregulate the expression of pcna and col-IV, and delay the progression of diabetic nephropathy.
