Abstract
Bovine bone was prepared into bone mud as the test material, and the conditions for pretreatment degreasing and decalcification for extracting bovine collagen polypeptide were studied. Compare the degreasing effects of three degreasing agents: petroleum ether, absolute ethanol, and n-hexane; use quadratic orthogonal rotation combination design to optimize decalcification conditions based on single factor experiments. The results show that the optimal conditions for the degreasing process are absolute ethanol as the bone mud degreasing agent, bone mud and absolute ethanol material-liquid ratio 1:20 (g/mL), 150r/min vibration soaking for 24 hours, and the bone mud degreasing rate can reach 97.46%. The optimal conditions for the citric acid decalcification process of bone mud are citric acid concentration 0.43 mol/L, decalcification time 16 hours, material-liquid ratio 1:21 (g/mL), and the decalcification rate can reach 96.53%.
