Abstract
In order to study the optimal process for extracting anglerfish skin collagen, anglerfish skin was used as the research object, and a single factor test was conducted by selecting three variables: liquid-to-material ratio, enzyme amount and enzymatic hydrolysis time, and then based on the Box-Behnken central combination test The design principle is to use three-factor and three-level response surface analysis (RSM) on the basis of a single factor to prepare pepsin-soluble collagen (PSC) and analyze its physical and chemical properties. The results show that the optimal extraction process conditions for anglerfish skin collagen are 4¡ãC, liquid-to-material ratio 18:1, 3% enzyme addition, and 48 hours of extraction time. Under these conditions, the extraction rate of anglerfish skin collagen is 2.11%; amino acid analysis The Arg and Tyr contents in the fish skin were 16% and 18% respectively; differential scanning calorimeter (DSC) measured the heat shrinkage temperature of PSC as 48.9¡ãC; SDS-PAGE electrophoresis showed that collagen basically retained the triple helix structure; electron Microscope (SEM) scan shows that PSC has a porous network structure
