Abstract
Using grass carp scales as raw materials, the acid-soluble collagen (ASC) and pepsin-soluble collagen (PSC) in the fish scales were extracted respectively. Based on the characterization of the collagen molecular structure, the in vitro self-assembly behavior of fish scale collagen was studied. And compared with porcine skin collagen (PC) of mammalian origin. The results showed that the three types of collagen prepared were all typical type I collagen. The molecular weight and secondary structure of the three types of collagen were relatively similar, but there were certain differences in the amino acid composition. The turbidity experiment was used to observe the in vitro self-assembly behavior of collagen. The results showed that under appropriate temperature and pH conditions, all three types of collagen can produce in vitro self-assembly behavior. The self-assembly process can be divided into hysteresis segment, There are three stages: rapid rising stage and stable stage, and their appearance time is affected by the pH value of the solution; under the same conditions, PC has the highest degree of self-assembly in vitro, followed by fish scale PSC and ASC; ambient temperature, collagen mass concentration and protein Thermal denaturation treatment will affect the in vitro self-assembly behavior of collagen to varying degrees.
